The Tankyrase 1 Activity Assays are high-throughput, semi-quantitative ELISAs designed to measure Tankyrase 1 activity. Tankyrase 1, also known as PARP5a, is a Poly (ADP-ribose) Polymerase (PARP) that associates with telomeres via its interaction with the telomeric-repeat binding factor TRF1. These assays utilize a histone-coated 96-well microplate and an anti-poly (ADP-ribose) (PAR) antibody to detect PAR that has been deposited onto the immobilized histone proteins by Tankyrase 1. Assays are available in a colorimetric detection format, which uses TACS-Sapphire™, or a chemiluminescent detection format, which uses PARP PeroxyGlow™ A and B. Additionally, these are the only commercially available assays to feature a full length Tankyrase 1 enzyme and unmodified NAD substrate to create a more physiologically applicable in vitro assay system.
